This often requires regeneration of the membrane by stripping off the antibodies and reprobing 19. However, chemiluminescence is not amenable to multi-target detection when using antibodies with low specificity or for proteins that are very close in molecular weight with overlapping signals. Multiplexed target detection from chemiluminescent western blots is feasible when working with highly specific antibodies against targets that differ in molecular weight by at least five to ten kilodaltons (kDa). With limiting sample availability to assess components of microdissected tissue, sorted cell subpopulations, exosomes and primary cells cultured from small tissue specimens, the requirement to maximize quantity while preserving the quality of data is paramount 17, 18. However, a complete and concise guide to the workflow highlighting the importance of reagent selection, data workup and analysis remains elusive. These highly viewed and well cited articles support the need for updated guidance in producing quantitative western blot data. Recent articles have dissected and contrasted the old practices passed down from the early days of western blotting to offer revised methodologies that take full advantage of the currently available state-of-the-art instruments and reagents to achieve accurate, precise and reproducible data 14, 15, 16. The associated retractions from high profile articles 10, 11, 12, 13 support the unfounded reputation of western blotting as a difficult, frustrating and non-reproducible technique that is semi-quantitative at best. However, the original qualitative techniques and assumptions that have been passed down through generations of lab members were not formally updated to permit the production of semi-quantitative western blot data thus giving poor reproducibility and misleading interpretations 7, 8, 9. This permitted the generation of semi-quantitative data from chemiluminescence. During this time, the first camera-based imaging systems were introduced which enabled automated detection over a much broader linear dynamic range of about three to four orders of magnitude 6. Since the linear quantitative range of film is only about an order of magnitude and very challenging to assess saturation by eye, the results even between technical replicates were difficult to reproduce 5. Over the following two decades, as reagent and antibody technologies evolved, scientists attempted to produce quantitative results initially by scintillation counting and film densitometry 3, 4. Western blotting was first introduced in 1979 as a means of detecting a target protein in a sample with antibodies and detection using film 1, 2. The data reveal when and how to apply these detection methods to achieve reproducible data with a stepwise approach to data processing for quantitative analysis. A methodology to optimize and produce excellent, quantitative western blot results with rigorous data analysis from membranes probed with both fluorescent and chemiluminescent antibodies is described. Fluorescence reagent and detection solutions are the latest innovation in western blotting but there remains questions and debate concerning their relative sensitivity and dynamic range versus chemiluminescence. The development of sensitive, camera-based detection technologies coupled with an updated technical approach permits the production of reproducible, quantitative data. Chemiluminescence coupled with film-based detection was traditionally the gold standard for western blotting but accurate and reproducible quantification has been a major challenge from this methodology. Western blotting experiments were initially performed to detect a target protein in a complex biological sample and more recently, to measure relative protein abundance. ![]() ![]() ![]() You rolled your eyes as you sat down in a chair trying your hardest not to smile. However, using a separate RBI and compass, this requires considerable mental calculation to determine the appropriate relative bearing. When the needle reaches an RBI reading corresponding to the required bearing, then the aircraft is at the position. An NDB may also be used to locate a position along the aircraft's current track (such as a radial path from a second NDB or a VOR). Having determined the drift, the aircraft must be flown so that the compass heading is the required bearing adjusted for drift at the same time as the RBI reading is 0 or 180 adjusted for drift. To do this it is necessary to correlate the RBI reading with the compass heading. When tracking to or from an NDB, it is also usual that the aircraft track on a specific bearing. ![]() This gives the magnetic bearing that must be flown: (RB + MH) mod 360 = MB. The formula to determine the compass heading to an NDB station (in a no wind situation) is to take the relative bearing between the aircraft and the station, and add the magnetic heading of the aircraft if the total is greater than 360 degrees, then 360 must be subtracted. ![]() Aircraft heading +/- ADF needle degrees off nose or tail = Bearing to or from NDB station. With a crosswind, the needle must be maintained to the left or right of the 0 or 180 position by an amount corresponding to the drift due to the crosswind. Similarly, the aircraft will track directly away from the NDB if the needle is maintained on the 180 degree mark. The aircraft will then fly directly to the NDB. In order to track toward an NDB (with no wind), the aircraft is flown so that the needle points to the 0 degree position. This display looks like a compass card with a needle superimposed, except that the card is fixed with the 0 degree position corresponding to the centreline of the aircraft. This bearing may be displayed on a relative bearing indicator (RBI). The ADF can also locate transmitters in the standard AM medium wave broadcast band (530 kHz to 1700 kHz at 10 kHz increments in the Americas, 531 kHz to 1602 kHz at 9 kHz increments in the rest of the world).ĪDF equipment determines the direction or bearing to the NDB station relative to the aircraft by using a combination of directional and non-directional antennae to sense the direction in which the combined signal is strongest. NDB navigation consists of two parts - the automatic direction finder (ADF) equipment on the aircraft that detects an NDB's signal, and the NDB transmitter. Main article: Automatic direction finder Automatic direction finder (ADF) equipment points to the direction of an NDB. The last two types are used in conjunction with an instrument landing system (ILS).Īutomatic direction finder equipment There are four types of non-directional beacons in the aeronautical navigation service: Non-directional beacons in North America are classified by power output: "low" power rating is less than 50 watts "medium" from 50 W to 2,000 W and "high" at more than 2,000 W. In Canada, privately owned NDB identifiers consist of one letter and one number. Each NDB is identified by a one, two, or three-letter Morse code callsign. NDBs used for aviation are standardised by International Civil Aviation Organization (ICAO) Annex 10 which specifies that NDBs be operated on a frequency between 190 kHz and 1750 kHz, although normally all NDBs in North America operate between 190 kHz and 535 kHz. The system, developed by United States Air Force (USAF) Captain Albert Francis Hegenberger, was used to fly the world's first instrument approach on May 9, 1932. However, NDB signals are also affected more by atmospheric conditions, mountainous terrain, coastal refraction and electrical storms, particularly at long range. NDB signals follow the curvature of the Earth, so they can be received at much greater distances at lower altitudes, a major advantage over VOR. NDB are in contrast to directional radio beacons and other navigational aids, such as low-frequency radio range, VHF omnidirectional range (VOR) and tactical air navigation system (TACAN). Radio beacons are radio transmitters at a known location, used as an aviation or marine navigational aid. A hollow square superimposed on this symbol indicates a collocated distance measuring equipment (DME) installation.Ī non-directional beacon ( NDB) or non-directional radio beacon is a radio beacon which does not include inherent directional information. I'm slowly reading Proust's À la recherche du temps perdu or In Search of Lost Time or whatever the current favored translation of the title is, and by "slowly" I mean I am still in the first part, which is called " SWANN's Way" ( 20D: Titular protagonist in a Marcel Proust novel).
